1.0 OBJECTIVE
To provide a guideline for operation, cleaning and calibration procedure of NUVE Bench Top centrifuge machine.
2.0 SCOPE
This SOP is applicable for the operation, cleaning and calibration procedure of NUVE Bench Top centrifuge machine at Quality Control Department (Company Name).
3.0 RESPONSIBILITY
3.1 Operating personnel in the laboratory is responsible for carrying out the procedure.
3.2 Overall responsibility for training, implementation & follow-up with the Head of QC or his/her nominee.
4.0 ACCOUNTABILITY
4.1 Head of Quality.
5.0 PRECAUTIONS
5.1 The emergency lid release must not be operated while the motor is running.
5.2 Operation of the centrifuge in explosion-hazardous location is prohibited.
5.3 The centrifuge should never be operated if :
-a breakdown exists or defective accessories are inserted.
-the rotor is not installed exactly.
-the rotor is not loaded symmetrically.
5.4 In case of imbalance (e.g. tube breakage or non-symmetrical loading) press stop key otherwise the centrifuge could be damaged.
5.5 Do not misuse the protective containers as sample vessels.
6.0 PROCEDURE
6.1 Set up
6.1.1 Connect the plug of the centrifuge to the main power.
6.1.2 Press the lid key to open the lid.
6.1.3 Fill the centrifuge tubes with the required volume of the sample solution.
6.1.4 Put the tubes into the protective containers and insert them completely into the rotor boring symmetrically.
6.1.5 For one centrifuge tube put another centrifuge tube in opposite rotor position filled with equal volume of liquid (usually water).
6.1.6 After loading close the lid.
6.1.7 Press the Function key to set the speed. By pressing up and down key set the desired speed (500 rpm-14000 rpm) Press the Function key again to register the speed.
6.1.8 Again press the Function key to set the time. By pressing up and down key set the desired time (1 min-99 min) Press the Function key again to register the time.
6.1.9 Again press the Function key to set the Acceleration and Break rate. By pressing up and down key set the desired rate (0-9) Press the Function key again to register the Acceleration and Break rate.
6.1.10 Press start key to run the centrifuge machine.
6.1.11 After the desired set time, the machine will stop automatically. After sometimes machine will beep and lid indicator light up.
6.1.12 The operation can be terminated by pressing the stop key.
6.2 Operation logbook
6.2.1 Record the informations in the operational log book- “LOG BOOK OF CENTRIFUGE MACHINE” as per FRM No.: FQC/446.
6.3 Cleaning
6.3.1 To clean the plastic surface a cloth moistened with non-corrosive liquid should be used.
6.3.2 If a glass or plastic tube breaks, the glass splinters and the spilled liquid should be removed carefully.
6.4 Calibration
6.4.1 Ensure that the instrument is clean before use, including surrounding area.
6.4.2 Check and ensure due date of calibration.
6.4.3 Operate the Centrifuge tester as per procedure.
6.4.4 Switch ON the power supply.
6.4.5 Set the time 5, 10 and 20 minutes by selecting TIMER key and start the machine and calibrated stopwatch simultaneously.
6.4.6 Note the actual time shown by a calibrated stopwatch.
6.4.7 Set a rotor of RS 24 acili.
6.4.8 Set 1000, 5000 and 10,000 rpm by Function key and start the instrument.
6.4.9 Record the number of rotation per minute.
6.4.10 Record the observations in the Calibration report (Annexure –II).
6.4.11 After completion of calibration switch ‘OFF’ the main supply.
6.4.12 If the acceptance criteria for calibration are not satisfied, then follow the procedure for Handling of Out of calibration for instrument & equipment (SOP No.: QA049).
6.4.13 After completion of the calibration activity, affix the duly filled and signed calibration status label on the instrument.
6.5 Calibration Frequency
6.5.1 Every year ± 10 days of due date and after maintenance job.
